报告内容

Copper-mediated trifluoromethylation represents one of the most efficient methods that may have potentially applications in the preparation of trifluoromethylated drug molecules. Mechanistically, it was proposed that the reaction proceeds via a Cu(I)/Cu(III) catalytic cycle involving two key intermediates [CuCF3] and [Ar-CuIII-CF3]. Nevertheless, both the Cu(I) species [CuCF3] and the putative key product-forming Cu(III) intermediate [Ar-CuIII-CF3] have never been observed or isolated previously. We now report the synthesis, characterization of both intermediates including stable Cu(I) species [nBu4N][Cu(CF3)2] and aryl(tris(trifluoromethyl))cuprate(III) complexes [nBu4N][Cu(Ar)(CF3)3]. Mechanistic investigations including kinetic studies, the effect of temperature and solvents, the effect of the para-substitution of the aryl group and DFT calculations suggest that C(sp2)-CF3 reductive elimination from [nBu4N][Cu(Ar)(CF3)3] proceeds via a concerted carbon-carbon bond-forming pathway.
